Why are US and imperial MPG different?

The gallon itself differs: a US gallon is 3.785 L while an imperial gallon is 4.546 L. The same car shows a bigger MPG number in the UK purely from the bigger unit — 40 imperial MPG is only about 33 US MPG.

How do I convert MPG to L/100 km?

Divide 235.215 by the US MPG figure (282.481 for imperial). It is a reciprocal relationship, so improving from 20 to 25 MPG saves far more fuel per mile than going from 40 to 45 — L/100 km makes that obvious.

Which unit is 'better'?

L/100 km is fuel per fixed distance, so consumption adds up linearly and comparisons are honest. MPG rewards already-efficient cars in misleading ways — which is why most of the world, and now plug-in hybrid ratings, use litres (or kWh) per distance.
